Abstract: Using strychnine as the template, methacrylic acid (MAA) as the functional monomer, ethylene glycol dimethacrylate ( EDMA) as the cross-linker, a strychnine imprinted monolithic column was prepared for the separation of the strychnine and brucine by in-situ, molecular imprinted technique. Then, the synthesis condition was optimized. The result showed the best ratio of template, functional monomer and cross-link was 1:4:16, and the toluene content in porogen was 18%. The effects of mobile phase composition, flow rate and column temperature on retention and separation factor were investigated. The maximum separation factor between strychnine and brucine was 3.5
