Abstract: The preparation is described of a cinchonine-imprinted monolithic polymer directly in a capillary column by in-situ polymerization before covalently linking to the activated inner surface of such column. Optimization of the reaction conditions to enhance polymer permeability and pore sizes was studied as well. The column was used in the separation of diastereomers of cinchonine and cinchonidine by pressure-assisted capillary electrochromatography with low electroosmotic flow in operation. Effects of applied voltage, pressure, temperature, pH value of mobile phase and its acetonitrile content on retention time and resolution were investigated. The separation took only 2 min.
