Abstract: This chapter discusses chiral separation of active pharmaceutical ingredients (APIs) and their related compounds by various methods. Particular emphases have been put on chiral separation by HPLC on chiral stationary phases (CSPs). Different types of CSPs, including polysaccharide derivatives, macrocyclic glycopeptides (antibiotics), cyclodextrins, Pirkletype, proteins, ligand exchange, crown ethers, synthetic polymers/molecularly imprinted polymers (MIPs), are discussed. Separation mechanisms and method development for chiral molecules using these phases are also discussed
