Abstract: The ampicillin (AMPI) molecularly imprinted polymer microspheres (MIPMs) via aqueous suspension polymerization were synthesized for the first time. Evaluation on the MIPMs by chromatographic analysis indicated that the optimum type and ratio of functional monomer to the template was 2-(Diethylamino) ethyl methacrylate (DEAEM) and 8:1, respectively. Under the optimized MISPE conditions, the recoveries were all above 89% for spiked AMPI in honey samples. After MISPE treatment, the obtained AMPI chromatograms showed that the matrix interferences could be almost neglected. The results demonstrate that the obtained MIPMs are applicable as inner adsorbents in MISPE cartridges for extraction of AMPI in foods
