Abstract: A molecularly imprinted monolithic column with special molecule recognition ability for melamine (MAM) was prepared by in situ polymerization method as the HPLC stationary phase. MAM was used as template molecule, methacrylic acid (MAA) as functional monomer and ethylene glycol dimethacrylate (EDMA) as cross-linker during the polymerization process. Pulsed elution format was applied to rapid screening elution agent and frontal chromatography method also applied to investigate the binding capacities of imprinted and non-imprinted monoliths. The experimental results showed that the imprinted monolithic column possesses good affinity to MAM. MAM can be thoroughly reserved by the imprinted column. The numbers of binding sites and the dissociation constants were determined as 2.28 x 103 μmol/g and 2.45 x 10 -5 mol/L for imprinted column, 366 μmol/g and 23.7 mol/L for non-imprinted column, respectively
