Abstract: The focus of this chapter is on the development of various monoliths for sample preparation, including polymer monoliths, silica monoliths, and organic-silica hybrid monoliths. The properties of various monolithic materials and the application fields are discussed. Furthermore, chemical modifications of the monolith surface and its effects on sample pretreatment efficiency are reviewed. The applications of monolithic materials for biomolecule analysis, drug analysis, and environmental sample analysis are discussed, with significant focus on the hyphenation of monolith-based sample preparation with separation and detection techniques. Furthermore, the potential trend for the development of multi-dimensional analytical systems with online monolith-based microextraction techniques is discussed
