The invention discloses a preparation method of amino modified magnetic molecularly imprinted polymer micro-spheres of chloramphenicol. The preparation method comprises the steps as follows: adding magnetic nanometer ferroferric oxide particle modified via oleic acid to a dispersing agent; then sequentially adding polymerizing monomer, functional monomer and cross-linking agent; ultrasonically dispersing; adding initiator; heating; agitating to react; magnetically separating; washing until pH (Potential of Hydrogen) is of 6 to 8; drying; adding the composite reacting liquid of chloramphenicol and the amino functional agent; heating; agitating to react; magnetically separating; washing until the pH is of 6 to 8; adding the organic solvent; ultrasonically eluting to remove the chloramphenicol until no chloramphenicol is detected via quick high performance liquid chromatography-tandem mass spectrum; and then drying to obtain the target product. The preparation method disclosed by the invention has the characteristics of being simple in preparation technology, and controllable in magnetic content and functional group ratio; and the preparation method can be used for enriching the trace chloramphenicol residual in the animal original free food to achieve high enrichment factor.
